Hello, this may seem like a stupid question, do I need to remove the spindle nuts to remove the rear rotors on my 2007 Honda Accord V6?

Nope. Maybe they're stuck on with corrosion. Use PB Blaster & a hammer.
There's probably a couple threaded holes in rotor, in between the wheel studs. Find a couple bolts that fit (M6??) & use them to drive the rotor off of the hub.
